Microbial dyes: dyeing of poplar veneer with melanin secreted by Lasiodiplodia theobromae isolated from wood
Abstract Dyeing methods that conform to environmental protection requirements and adapt to the development of industry are investigated using bleached poplar wood dyed with melanin secreted by Lasiodiplodia theobromae in this paper. Prolonging the dyeing time of coarse melanin can improve the dye uptake of veneers, and the brightness value increases, and the reflectivity declines with the increase of dyeing time. The stained surface of the vessel is gradually covered by coarse...

Metabolomic change and pathway profiling reveal enhanced ansamitocin P-3 production in Actinosynnema pretiosum with low organic nitrogen availability in culture medium
Abstract Ansamitocin P-3 (AP-3), a 19-membered polyketide macrocyclic lactam, has potent antitumor activity. Our previous study showed that a relatively low organic nitrogen concentration in culture medium could significantly improve AP-3 production of Actinosynnema pretiosum. In the present study, we aimed to reveal the possible reasons for this improvement through metabolomic and gene transcriptional analytical methods. At the same time, a metabolic pathway profile based on...

Stepwise optimization of recombinant protein production in Escherichia coli utilizing computational and experimental approaches
Abstract Over the past few decades, Escherichia coli (E. coli) remains the most favorable host among the microbial cell factories for the production of soluble recombinant proteins. Recombinant protein production (RPP) via E. coli is optimized at the level of gene expression (expression level) and the process condition of fermentation (process level). Presently, the reported studies do not give a clear view on the selection of methods employed in the optimization of RPP. Here,...

A novel DNA methylation motif identified in Bacillus pumilus BA06 and possible roles in the regulation of gene expression
Abstract Single-molecule real-time (SMRT) sequencing can be used to identify a wide variety of chemical modifications of the genome, such as methylation. Here, we applied this approach to identify N6-methyl-adenine (m6A) and N4-methyl-cytosine (m4C) modification in the genome of Bacillus pumilus BA06. A typical methylation recognition motif of the type I restriction-modification system (R-M), 5′-TCm6AN8TTGG-3′/3′-AGTN8m6AACC-5′, was identified. We confirmed that this motif was...

Development of a highly efficient and specific l -theanine synthase
Abstract γ-Glutamylcysteine synthetase (γ-GCS) from Escherichia coli, which catalyzes the formation of l-glutamylcysteine from l-glutamic acid and l-cysteine, was engineered into an l-theanine synthase using l-glutamic acid and ethylamine as substrates. A high-throughput screening method using a 96-well plate was developed to evaluate the l-theanine synthesis reaction. Both site-saturation mutagenesis and random mutagenesis were applied. After three rounds of directed evolution,...

Alginate oligosaccharide improves lipid metabolism and inflammation by modulating gut microbiota in high-fat diet fed mice
Abstract Alginate oligosaccharides are associated with some beneficial health effects. Gut microbiota is one of the most recently identified factors in the development of several metabolic diseases induced by high-fat diet. Our objective was to evaluate how alginate oligosaccharides impact on high-fat diet‑induced features of metabolic disorders and whether this impact is related to modulations in the modulation of the gut microbiota. C57BL/6J mice were fed with chow diet, high-fat...

Perspectives for using glacial and periglacial microorganisms for plant growth promotion at low temperatures
Abstract Even though they are among the most extreme environments in which life can develop, glaciers are colonized by metabolically active microbes, some of which thrive—in their own particular way—under the prevailing harsh conditions. Glacial or periglacial microbes are often psychrophiles since they are able to grow optimally at low temperatures. This ability has evolved through a series of adaptations, both molecular and physiological, some of which have been exploited...

Bacterial non-specific nucleases of the phospholipase D superfamily and their biotechnological potential
Abstract Bacterial non-specific nucleases are ubiquitously distributed and involved in numerous intra- and extracellular processes. Although all nucleases share the basic chemistry for the hydrolysis of phosphodiester bonds in nucleic acid molecules, the catalysis comprises diverse modes of action, which offers great potential for versatile biotechnological applications. A major criterium for their differentiation is substrate specificity. Specific endonucleases are widely used...

Endometriosis in pregnancy
Abstract Endometriosis constitutes the presence of ectopic endometrial glands and stroma outside the uterine endometrium, which is hormonally responsive and responds to pregnancy hormones as well. Decidualization is a physiologic process, where the normal endometrium readies itself for optimal accommodation of a pregnancy. A similar hormonal response can be seen with ectopic endometrium as well. As such, ovarian endometriomas and deep endometriosis implants can undergo decidualization....
