Roles of active site residues in catalysis, substrate binding, cooperativity and the reaction mechanism of the quinoprotein glycine oxidase [Enzymology]
The quinoprotein glycine oxidase from the marine bacterium Pseudoalteromonas luteoviolacea (PlGoxA) uses a protein-derived cysteine tryptophylquinone (CTQ) cofactor to catalyze conversion of glycine to glyoxylate and ammonia. This homotetremeric enzyme exhibits strong cooperativity towards glycine binding. It is a good model for studying enzyme kinetics and cooperativity, specifically for being able to separate those aspects of protein function through directed mutagenesis. Variant proteins were...

The single CCA-adding enzyme of T. brucei has distinct functions in the cytosol and in mitochondria [RNA]
tRNAs universally carry a CCA nucleotide triplet at their 3′-ends. In eukaryotes, the CCA is added post-transcriptionally by the CCA-adding enzyme (CAE). The mitochondrion of the parasitic protozoan Trypanosoma brucei lacks tRNA genes and therefore imports all of its tRNAs from the cytosol. This has generated interest in the tRNA modifications and their distribution in this organism, including how CCA is added to tRNAs. Here, using a BLAST search for genes encoding putative CAE proteins in T. brucei,...

Structure and allosteric activity of a single disulfide conopeptide from Conus zonatus at human {alpha}3{beta}4 and {alpha}7 nicotinic acetylcholine receptors [Protein Structure and Folding]
Conopeptides are neurotoxic peptides in the venom of marine cone snails and have broad therapeutic potential for managing pain and other conditions. Here, we identified the single-disulfide peptides Czon1107 and Cca1669 from the venoms of Conus zonatus and Conus caracteristicus, respectively. We observed that Czon1107 strongly inhibits the human α3β4 (IC50 15.7 ± 3.0 μM) and α7 (IC50 77.1 ± 0.05 μM) nicotinic acetylcholine receptor (nAChR) subtypes, but the activity of Cca1669 remains to be identified....

A flexible network of Vimentin intermediate filaments promotes the migration of amoeboid cancer cells through confined environments [Cell Biology]
Tumor cells can spread to distant sites through their ability to switch between mesenchymal and amoeboid (bleb-based) migration. Because of this difference, inhibitors of metastasis must account for each migration mode. However, the role of Vimentin in amoeboid migration has not been determined. Since amoeboid, Leader Bleb-Based Migration (LBBM) occurs in confined spaces and Vimentin is known to strongly influence cell mechanical properties, we hypothesized that a flexible Vimentin network is required...

MAFB promotes cancer stemness and tumorigenesis in osteosarcoma through a Sox9-mediated positive feedback loop
Despite the fact that osteosarcoma is one of the most common primary bone malignancies with poor prognosis, the mechanism behind the pathogenesis of osteosarcoma is only partially known. Here we characterized differentially expressed genes (DEG) by extensive analysis of several publicly available gene expression profile datasets and identified MAFB as a key transcriptional regulator in osteosarcoma progression. MAFB was highly expressed in tumor tissues and required for proliferation and tumorigenicity...

Patient and surgery factors associated with the incidence of failed and difficult intubation
Summary Estimates of the rate and risk‐factors for difficult airway rarely include a denominator for the number of anaesthetics. Approaches such as self‐reporting and crowd‐sourcing of airway incidents may help identify specific lessons from clinical episodes, but the lack of denominator data, biased reporting and under‐reporting does not allow a comprehensive population‐based assessment. We used an established state‐wide dataset to determine the incidence of failed and difficult intubations between...

Bullous pemphigoid and dipeptidyl peptidase-4 inhibitors: a meta-analysis of randomized controlled trials
Abstract Purpose An increasing body of evidence suggests that dipeptidyl-peptidase 4 (DPP-4) inhibitors could play a role in the development of bullous pemphigoid. The knowledge regarding this association is based on case reports, pharmacovigilance database analyses, and observational studies. Data from randomized clinical trials are a relevant source of information on adverse events. Since no single trial has a sufficient power to...

The calcium-to-phosphorous (Ca/P) ratio in the diagnosis of primary hyperparathyroidism and hypoparathyroidism: a multicentric study
Abstract Purpose The diagnosis of primary hyperparathyroidism (PHPT) and chronic hypoparathyroidism (HypoPT) is still challenging, especially in patients asymptomatic or with non-classical phenotypes and for physicians not skilled in calcium-phosphorous (Ca–P) disorders. The serum calcium/phosphorous (Ca/P) ratio has been proposed as accurate index to identify PHPT, while it has never been tested in HypoPT. The aim of this study is...

Survival benefits of simple versus extended cholecystectomy and lymphadenectomy for patients with T1b gallbladder cancer: An analysis of the surveillance, epidemiology, and end results database (2004 to 2013)
Our study demonstrated that using extensive lymphadenectomy (≥ 5 lymph nodes) would provide a significant survival benefit. The substantial effort is recommended to made to increase the use of lymphadenectomy, radical resection/radical cholecystectomy, and extended surgical resection in the treatment of gall bladder cancer. However, the survival benefit of extended surgical resection was not significantly better than that of simple cholecystectomy, and could have been affected by few patients who...

Pemetrexed sensitizes human lung cancer cells to cytotoxic immune cells
Abstract Pemetrexed (PEM) is a useful drug that can be combined with immune checkpoint blockade therapy for treatment of patients with advanced non‐small‐cell lung cancer (NSCLC). However, its effects on anti‐cancer immunity, especially the sensitivity of NSCLC cells to cytotoxic immune cells, have not been fully investigated. In this study, we examined the effects of PEM on the sensitivity of human NSCLC cells to two different types of cytotoxic immune cells. Pre‐treatment with PEM increased the...
